Oncostatin M (OSM) is a multifunctional cytokine that belongs to the Interleukin-6 subfamily. Among the family members, OSM is most closely related to leukemia inhibitory factor (LIF) and it in fact utilizes the LIF receptor in addition to its specific receptor in the human. A biologically active OSM receptor has been previously described that consists of a heterodimer of leukemia inhibitory factor receptor (LIFR) and gp130. OSM is synthesized by stimulated T-cells and monocytes. The effects of OSM on endothelial cells suggest a pro-inflammatory role for OSM and endothelial cells possess a large number of OSM receptors. Background

Leukemia inhibitory factor (LIF) is a member of Interleukin 6 family. This protein is mainly expressed in the trophectoderm of the developing embryo, with its receptor LIFR expressed throughout the inner cell mass. LIF has the capacity to induce terminal differentiation in leukemic cells. Its activities include the induction of hematopoietic differentiation in normal and myeloid leukemia cells, the induction of neuronal cell differentiation, and the stimulation of acute-phase protein synthesis in hepatocytes. LIF is used in mouse embryonic stem cell culture, because that removal of LIF pushes stem cells toward differentiation, but they retain their proliferative potential or pluripotency. It is also used in phase II clinical trial, which can assist embryo implantation in women who have failed to become pregnant despite assisted reproductive technologies (ART). Background

CD40 ligand is a 261 amino acid type II transmembrane glycoprotein belonging to the TNF family and can be cleaved into two chains: the CD40 ligand membrane form and CD40 ligand soluble form. It is primarily expressed on activated CD4+ T lymphocytes, and also found in other types of cells, like NK cells, mast cells, basophils and eosinophils. In total CD40 ligand has three binding partners: CD40, α5β1 integrin and αIIbβ3. It mediates a range of activities on B cells including induction of activation-associated surface antigen, entry into cell cycle, isotype switching and Ig secretion and memory generation. Background

Flt3-ligand (FL) is a recently identified hematopoietic cytokine whose activities are mediated by binding to the transmembrane glycoprotein Flt3. Flt3 was first discovered as a member of the class III subfamily of receptor tyrosine kinases (RTK) whose expression among hematopoietic cells was found to be restricted to highly enriched stem/progenitor cell populations. Additionally, class III RTKs include the receptors from SCF, M-CSF and PDGF. Not surprisingly, Flt3-ligand is also structurally related to M-CSF and SCF. All three cytokines have been shown to exist both as type I transmembrane proteins and as soluble proteins. The predominant human FL isoform is a transmembrane protein that can undergo proteolytic cleavage to generate a soluble form of the protein. Background

B Cell Activating Factor Receptor (BAFF-R), also named tumor necrosis factor receptor superfamily member 13C, is a member of the TNFR superfamily. It is highly expressed in spleen, lymph node, and resting B cells and to some extent in activated B cells, resting CD4+ cells and peripheral blood leukocytes. BAFF receptor is a type III transmembrane protein containing a single extracellular phenylalanine-rich domain and binds with high specificity to BAFF (TNFSF13B). It enhances B-cell survival in vitro and is a regulator of the peripheral B-cell population. BAFF receptor/BAFF signaling plays a critical role in B cell survival and maturation.

Background

B-cell activating factor (BAFF), also known as Blys, TALL-1, TNAK, and zTNF4, is a TNF ligand superfamily member and has been designated TNFSF13B. Produced by macrophages, dendritic cells, and T lymphocytes, BAFF promotes the survival of B cells and is essential for B cell maturation. BAFF binds to three TNF receptor superfamily members: B-cell maturation antigen (BCMA/TNFRSF17), transmembrane activator and calcium-modulator and cyclophilin ligand interactor (TACI/TNFRSF13B) and BAFF receptor (BAFF R/BR3/TNFRSF 13C). These receptors are type III transmembrane proteins that lack a signal peptide. Whereas TACI and BCMA bind BAFF and another TNF superfamily ligand, APRIL (a proliferation-inducing ligand), BAFF R selectively binds BAFF. TNF-beta, also known as lymphotoxin-alpha (LTalpha) and TNF-alpha are two structurally and functionally related proteins that bind to the same cell surface receptors (TNF RI and TNF RII) and produce a vast range of similar, but not identical effects. Background

The clinical use of the potent antitumor activity of TNF-α has been limited by the proinflammatory side effects including fever, dose-limiting hypotension, hepatotoxicity, intravascular thrombosis, and hemorrhage. Designing clinically applicable TNF-αmutants with low systemic toxicity has been an intense pharmacological interest. Background

Tumor necrosis factor alpha (TNF-α), also called cachectin, is the best-know member of the TNF-family, which can cause cell death. This protein is produced by neutrophils, activated lymphocytes, macrophages, NK cells, LAK cells, astrocytes endothelial cells, smooth muscle cells and some transformed cells. TNF-α occurs as a secreted, soluble form and as a membrane-anchored form, both of which are biologically active. The naturally-occurring form of TNF-α is glycosylated, but non-glycosylated recombinant TNF-α has comparable biological activity. The biologically active native form of TNF-α is reportedly a trimer. Human and murine TNF-α show approximately 79% homology at the amino acid level and cross-reactivity between the two species. Recombinant Human TNF-α contains soluble cytokines (77-233aa) of the protein. This protein was expressed in E.coli and purified without tag. It is expressed as soluble trimer, which has been validated by Size-exclusion chromatography. Apoptosis induced experiments confirmed that the protein had high biological activity.
TNF-α is a multifunctional proinflammatory cytokine that belongs to the tumor necrosis factor (TNF) superfamily,and it produced by various immune cells, including T cells, B cells, natural killer cells and macrophages. As a transmembrane protein, TNF-α’s membrane-bound site can be cleaved by the metalloprotease ADAM17/TACE, releasing the soluble cytokine into the extracellular space to form polymers [1-2]. The cellular response to TNF-α is mediated through the receptors TNFR1 and TNFR2. TNFR1 can be activated by both the membrane-bound and soluble trimeric forms of TNF-α, and resulting the downstream apoptosis signaling pathway. While the TNFR2 responds to the membrane-bound form of TNF-α, and promotes T cell activation and proliferation [3]. TNF-α plays a key regulatory role in inflammation and host defense against bacterial infection. The role of TNF-α in autoimmunity is underscored by blocking TNF-α action to treat rheumatoid arthritis and Crohn’s disease.

本产品可有效激活NF-κB的核转运信号通路,使JP6 (REL-associated protein, Rel-A)转移到细胞核内。稳定表达EGFP-JP6的HeLa细胞(C8001)经终浓度为10ng/ml的本产品刺激后EGFP-JP6进入细胞核,需要注意的是随着时间的延长EGFP-JP6会出核再次回到细胞质中,因此加入本产品后观察EGFP-JP6进入细胞核理想的时间为15-60分钟(图2)。

本产品经尺寸排阻层析(Size-exclusion chromatography, SEC)验证为三聚体。人体中TNF-α为26kDa的单次跨膜蛋白,TNF-α被TACE/ADAM17 (TNF-α Converting Enzyme/A Disintegrin and Metalloprotease 17)在Ala76-Val77之间切割,成为游离于细胞膜外可溶形式的TNF-α (77-233aa),无论是单次跨膜的TNF-α,还是游离于细胞膜外可溶形式的TNF-α,都以同源三聚体的构象行使其生物活性[4]。参考图4,本产品经16/60 Beyodex? 200pg验证为三聚体。

Background

Tumor necrosis factor alpha (TNF-α), also called cachectin, is the best-know member of the TNF-family, which can cause cell death. This protein is produced by neutrophils, activated lymphocytes, macrophages, NK cells, LAK cells, astrocytes endothelial cells, smooth muscle cells and some transformed cells. TNF-α occurs as a secreted, soluble form and as a membrane-anchored form, both of which are biologically active. The naturally-occurring form of TNF-α is glycosylated, but non-glycosylated recombinant TNF-α has comparable biological activity. The biologically active native form of TNF-α is reportedly a trimer. Human and murine TNF-α show approximately 79% homology at the amino acid level and cross-reactivity between the two species. Two types of receptors for TNF-α have been described and virtually all cell types studied show the presence of one or both of these receptor types.

Macrophage Colony Stimulating Factor (M-CSF), also named CSF-1, is a hematopoietic growth factor that is involved in the proliferation, differentiation, and survival of monocytes, macrophages, and bone marrow progenitor cells. It is produced by osteoblasts (as a result of endocrine stimulation by parathyroid hormone) exerts paracrine effects on osteoclasts and can interact with CSF1R. M-CSF is a four α-helical bundle cytokine and its active form is found extracellularly as a disulfide-linked homodimer. Four transcript variants encoding three different isoforms have been reported for M-CSF gene. Although forms may vary, all of them contain the N-terminal 150a.a. portion that is necessary and sufficient for interaction with the receptor. The first 223a.a. of mature human M-CSF shares 88%, 86%, 81% and 74% sequence identity with corresponding regions of dog, cow, mouse and rat M-CSF, respectively. Pigment epithelium-derived factor (PEDF) is encoded by the SERPINF1 gene in humans and found in verebrates. It is a secreted phosphoglycoprotein that belongs to the clade F subfamily, serpin superfamily of proteinase inhibitors. The PEDF is a noninhibitory serpin with neurotrophic, anti-angiogenic, and anti-tumorigenic properties. It is synthesized as a 418a.a. about 50kDa precursor that contains a 19a.a. signal sequence and a 399a.a. mature region that shows a pyroglutamate at Gln20. Like other serpins, it contains three β-sheets, 810 α-helices, and a C-terminal RCL (reactive center loop). Unlike other serpins with Ser protease inhibiting activity. PEDF has functions of inducing extensive neuronal differentiation in retinoblastoma cells, inhibiting of angiogenesis. As it does not undergo the S (stressed) to R (relaxed) conformational transition characteristic of active serpins, it exhibits no serine protease inhibitory activity. PEDF is researched as a therapeutic candidate for treatment of such conditions as choroidal neovascularization, heart disease, and cancer.

Background

Thymic stromal lymphopoietin (TSLP) is a hemopoietic protein belonging to the cytokine family and is known to play an important role in the maturation of T cell populations through activation of antigen presenting cells. It is mainly expressed in a number of tissues including heart, liver and prostate. TSLP signals through a heterodimeric receptor complex composed of the thymic stromal lymphopoietin receptor and the IL-7R alpha chain. After binding STAT5 phosphorylation is induced resulting in the expression of downstream transcription factors. Like IL-7, TSLP induces phosphorylation of STAT3 and STAT5, but uses kinases other than the JAKs for activation. TSLP has the functions of enhancing the maturation of CD11c+ dendritic cells and inducing allergic inflammation by directly activating mast cells. It expression is linked to many disease states including asthma, inflammatory arthritis, atopic dermatitis, and eczema and other allergic states. But the factors inducing the activation of TSLP release are not clearly defined. Human TSLP shares approximately 43%a.a. sequence identity with mouse TSLP.

Background

Granulocyte-Macrophage Colony Stimulating Factor (GM-CSF) is secreted by a number of different cell types (including activated T cells, B cells, macrophages, mast cells, endothelial cells and fibroblasts) in response to cytokine or immune and inflammatory stimulation. It was initially characterized as a growth factor that can support the in vitro colony formation of granulocyte-macrophage progenitors and has functions of stimulates the growth and differentiation of hematopoietic precursor cells from various lineages. GM-CSF has also been reported to have a functional role on non-hematopoietic cells and can induce human endothelial cells to migrate and proliferate. Additionally, it can stimulate the proliferation of a number of tumor cell lines, including osteogenic sarcoma, carcinoma and adenocarcinoma cell lines. Human GM-CSF shares 54% sequences identity with mouse GM-CSF, but has no biological effects across species. GM-CSF is used as a medication to stimulate the production of white blood cells following chemotherapy and has also recently been evaluated in clinical trials for its potential as a vaccine adjuvant in HIV-infected patients.

Background

Granulocyte colony stimulating factor (G-CSF) is a pleiotropic cytokine. It is mainly produced by monocytes and macrophages upon activation by endotoxin, TNF-α and IFN-γ. Besides, many other cell types can secreted this protein after LPS, IL-1 or TNF-α activation, which are fibroblasts, endothelial cells, astrocytes and bone marrow stromal cells. Various carcinoma cell lines and myeloblastic leukemia cells can express G-CSF constitutively. G-CSF is cytokine that acts in hematopoiesis by controlling the production, differentiation, and function of 2 related white cell populations of the blood, the granulocytes and the monocytesmacrophages. In addition it may function in some adhesion or recognition events at the cell surface.
In humans, two distinct cDNA clones for G-CSF, encoding 207 and 204 amino acid (a.a.) precursor proteins, have been isolated. Both proteins have a 30a.a. signal peptide and have identical amino acid sequences except for a threea.a. insertion (deletion) at the 35tha.a. residue from the N-terminus of the mature protein. Human G-CSF is 73% identical at the amino acid level to murine G-CSF and the two proteins show species cross-reactivity.

Stem Cell Factor (SCF) that binds to the c-Kit receptor is produced by fibroblasts and endothelial cells. The soluble and transmembrane forms of the protein are formed by alternative splicing of the same RNA transcript and the presence of both soluble and transmembrane SCF is required for normal hematopoietic function. SCF plays an important role in hematopoiesis, spermatogenesis, and melanogenesis. It also promotes mast cell adhesion, migration, proliferation, and survival. Background

Interleukin-36 receptor antagonist (IL-36RA) is a secreted protein which belongs to the interleukin 1 cytokine family (IL-1 family) and it is predominantly expressed in keratinocytes but not in fibroblasts, endothelial cells or melanocytes. IL-36RA has been reported to antagonize the biological activity of IL-36α (IL-1F6), IL-36β (IL-1F8), and IL-36γ (IL-1F9). Furthermore, it could constitute part of an independent signaling system analogous to interleukin-1 alpha (IL-1A), beta (IL-1B) receptor agonist and interleukin-1 receptor type I (IL-1R1).
