嘌呤霉素氨基核苷(Puromycin Aminonucleoside, PANS)是抗生素嘌呤霉素的氨基核苷部分,这一嘌呤霉素类似物不能抑制蛋白合成或诱导凋亡,替代的是用于诱导肾病的实验动物模型,以研究局灶节段性肾小球硬化、肾病变的诱导和引发蛋白尿症的事件。大鼠体内注射PANS(0.5-5 μM)能影响肾小球上皮细胞黏附,降低α3β1整合素表达。PANS还能减少肾病蛋白(Nephrin)和足突蛋白(Podocin)的表达。

实验方法(Assay):The 56/10 A1 cells were seeded at 0.01 × 106/cm2in 96-well plates and incubated for 2 d at 37℃. PAN was added to the media at a concentration of 0, 0.5, or 5.0 μg/ml (n = 12 for each PAN concentration), and the cells were incubated for another 48 h. Then the effect of PAN on proliferation of cells was assessed by an enzyme-linked immunosorbent assay method, using BrdU.

实验结果(Results):Control wells that were not exposed to PAN contained 204.3 ± 31.1 × 103cells/cm2, compared with 204.0 ± 30.6 × 103cells/cm2in wells that were exposed to 0.5 μg/ml PAN and 163.7 ± 36.3 × 103cells/cm2in wells that were exposed to 5.0 μg/ml PAN.

动物模型(Animal Model):Eight-week-old maleSprague-Dawley (SD) rats (weighing170–200g)

药物配制(Preparation):Puromycin aminonucleoside (PAN) was dissolved in dissolved in saline

实验方法(Assays):Rats were randomly divided into four groups: Control group; a PAN-treated Nephrotic syndrome (NS) group; a NS plus Losartan (30 mg/kg/day) treated group; and a NS plus PAL (200 mg/kg/day) treated group. Rats were intravenously injected with PAN (75 mg/kg/day, dissolved in saline). The control group was injected with saline only. Rats were administered either distilled water (Control and NS groups), Losartan or PAL by an oral gavage method once daily for 7 days.

实验结果(Results):Proteinuria is an important characteristic of puromycin aminonucleoside-induced NS in rats. 24-h urinary protein excretion progressively increased following PAN injection. Proteinuria levels in the NS group peaked on day 7 compared to the Control group. However, the PAL-treated group exhibited significantly decreased proteinuria levels on day 7 (p < 0.01). Similarly, treatment with losartan produced similar results to those seen in the PAL group.
